Glabridin can form a hydrogen bond with the active center of tyrosinase and firmly stuck at the entrance of tyrosinase, so that the raw material of melanin synthesis cannot enter and the semi-finished product of melanin cannot exit, reducing melanin production.

The cause of darkening of the skin in normal health is from tyrosinase and reactive oxygen species. Tyrosinase produces melanin on a regular basis, which changes the skin or eyes from brown to black.

Glabridin inhibits melanin synthesis by competitively inhibiting tyrosinase activity, taking away part of the tyrosinase from the catalytic ring of melanin synthesis and preventing the binding of substrates to tyrosinase, and also because glabridin itself has a good antioxidant effect. It has been proved that it is similar to superoxidedismutase (SOD) in scavenging oxygen free radicals and similar to vitamin E. 

It can inhibit both the activity of tyrosinase and dopa pigment interchange and dihydroxyindole carboxylic acid oxidase, which is a kind of fast, efficient and green cosmetic additive for whitening and spot removal. Some experimental data show that the whitening effect of glabridin is 232 times higher than that of ordinary vitamin C, 16 times higher than that of hydroquinone, and 1164 times higher than that of "arbutin".
